小菜蛾泛素基因的克隆与分析

Abstract:
运用rt-pcr技术,从小菜蛾幼虫总rna反转录产物中扩增出泛素基因的cdna序列.利用rna干扰(rnai)技术将一段体外合成的与小菜蛾泛素延伸蛋白基因uba52同源的dsrna注入四龄幼虫体内,以抑制小菜蛾uba52基因的表达.半定量rt-pcr结果显示,72h后沉默组小菜蛾幼虫体内uba52基因的mrna表达量显著下调.本实验成功沉默小菜蛾泛素基因,此rnai干扰体系的建立为利用此技术分析小菜蛾及其他昆虫基因的功能提供了参考.
