小菜蛾泛素基因的克隆及序列分析

Abstract:
设计了一对简并引物,从小菜蛾plutellaxylostella(l.)抗溴氰菊酯品系和敏感品系的雌雄成虫基因组中分别克隆了泛素基因的编码区,genbank登录号分别为eu28778,eu28779,eu28780,eu28781.序列分析表明,该基因的长度为228bp,编码的多肽由76个氨基酸组成.不同品系和不同性别的小菜蛾泛素基因的核苷酸序列、氨基酸序列等均存在差异,其中敏感品系雌雄成虫间差异较大,序列相似性为81.1%;抗性品系雌雄成虫间差异较小,序列相似性为97.8%.推测这些差异可能与小菜蛾抗药性的形成有关.因而为进一步研究小菜蛾抗药性形成机制和遗传机理奠定了基础.
