新月弯孢霉高产漆酶菌株的诱变及其培养条件优化

Abstract:

利用紫外线-60co-γ、微波及硫酸二乙酯诱变新月弯孢霉菌株,使其产漆酶能力大幅度提高。漆酶活性由原来的0.55μmol/(min?ml)提高到1.949μmol/(min?ml)。进一步通过单因素实验及正交实验得出最佳产酶条件为:发酵温度30℃,发酵时间72h,培养基ph为6,装液量120ml,摇床转速160r/min,土豆200g/l,蔗糖30g/l,大豆蛋白胨5g/l,kh2po45g/l,愈创木酚0.2mmol/l,吐温800.1ml/l,mg2+0.5mmol/l,cu2+1mmol/l,ca2+1mmol/l,mn2+0.25mmol/l。在此条件下,最终漆酶活性可达2.306μmol/(min?ml)。
