松材线虫携带的荧光假单胞菌pseudomonasfluorescensgcm5-1a菌株分泌的毒素及其对黑松的毒性

Abstract:

从松材线虫携带的致病细菌荧光假单胞菌pseudomonasfluorescensgcm5-1a菌株的培养液中分离、纯化得到一种50ku的蛋白质,其n-末端氨基酸顺序为alsvntnits,证明其为荧光假单胞标准菌株p.fluorescenspf-5的鞭毛蛋白相似,同时该蛋白能与pf-5菌株鞭毛蛋白抗体反应,证明其为相同的鞭毛蛋白。两个环二肽同时从gcm5-1菌株培养液中被分离和鉴定,它们是cyclo(-pro-val-)和cyclo(-pro-tyr-)。生测表明此次分离鉴定的鞭毛蛋白和两个环二肽对黑松愈伤组织及幼苗均具毒性。采用细胞免疫荧光分析、细胞染色观察、电导率测定、基因组dna电泳分析技术对上述鉴定的鞭毛蛋白与黑松愈伤组织的作用及致死方式进行了研究,结果表明:鞭毛蛋白与黑松细胞之间存在直接的互作;鞭毛蛋白处理的细胞其细胞膜皱缩变形、细胞质浓缩、细胞核变形,形成若干小核,细胞质中的rna降解;处理细胞着色力增强,细胞培养液的电导率增加,说明鞭毛蛋白科增加细胞膜的透性;基因组dna电泳分析证实处理细胞的dna发生断裂,但无明显的梯子形成,推测鞭毛蛋白对黑松细胞的致死方式为非典型的细胞凋亡。
