issr在油茶品种鉴别和遗传多样性分析中的应用

Abstract:

分子标记技术是准确鉴别油茶品种的重要手段之一。以湖南、湖北两省主要油茶品种为试验材料,利用issr分子标记技术分析了66个油茶品种的遗传多样性,揭示了各品种间的亲缘关系并对其进行了有效鉴别。结果表明:从100条引物中筛选出6条引物对66个油茶品种的基因组dna进行扩增,获得了121条可以统计的扩增产物,其中多态性条带数(npb)为112条,多态位点百分率为92.56%(ppb),平均等位基因观察值为1.9339,平均有效等位基因为1.6509,基因多样性指数即平均期望杂合度为0.3698,shannon信息指数为0.5417,说明油茶品种间的遗传多样性水平较高。根据扩增图谱条带的有无、多少和不同引物提供的谱带类型组合对供试油茶品种进行了鉴别。
