Abstract:

【目的】探寻脑红蛋白(Neuroglobin,NGB)在成年牦牛脑不同部位中的分布特征。【方法】选择健康成年牦牛8只,宰杀后取其大脑皮质、小脑皮质、海马、延髓、纹状体及嗅球等部位的脑组织,利用免疫组织化学染色SP法和实时荧光定量PCR技术,对NGB在成年牦牛脑不同部位中的分布特征及定量关系进行观察与检测。【结果】NGB在成年牦牛脑不同区域的表达强弱有明显差异,在大脑皮质中NGB基因的相对表达量(262.69±9.19)极显著高于小脑皮质(137.00±7.29)、海马(1.00±0.22)、延髓(3.43±0.76)、纹状体(7.65±0.61)及嗅球(2.14±1.22)。【结论】NGB在成年牦牛脑不同部位中高表达,提示NGB在脑组织的氧利用及功能活动中发挥着重要作用。
【Objective】This study aimed to explore the localization of neuroglobin (Neuroglobin,NGB) in different areas of adult yak brain.【Method】Eight health adult yaks were selected and NGB expression in different areas (cerebral cortex,cerebellar cortex,hippocampus,medulla oblongata,striaturn,and olfactory bulb) of adult yak brain was determined using the immunohistochemical SP technique and the real-time fluorescent quantitative PCR technology .【Result】 The expression of NGB in different areas of adult yak brain had significant difference.The relative expression of NGB gene in cerebral cortex (262.69±9.19) was significantly higher than in cerebellar cortex (137.00±7.29),hippocampus (1.00±0.22),medulla oblongata (3.43±0.76),striaturn (7.65±0.61),and olfactory bulb (2.14±1.22).【Conclusion】The expression of NGB in different areas of adult yak brain suggested that NGB might play an important role in utilization of oxygen and physiological functions
