不同浓度锰处理对甘草sqs1基因表达及其甘草酸含量影响的分析

Abstract:
以一年生甘草移栽苗为实验材料,设置5个锰离子浓度水平,分别为0、1.81、18.1、36.2和54.3mg·l-1(mnso4·h2o),利用实时荧光定量pcr和高效液相色谱方法对甘草鲨稀合成酶1(sqs1)基因相对表达量和甘草酸含量进行测定,从而探讨锰处理对甘草sqs1基因表达量与甘草酸积累的影响。结果表明随着锰处理浓度的增加,甘草sqs1基因的相对表达量和甘草酸含量均呈现先升高后下降的趋势,且两者之间存在极显著正相关(p<0.01,r=0.737)。sqs1基因表达量在18.1mg·l-1浓度处理时达到最大值为7.90,分别是对照(0mg·l-1)、1.81、36.2和54.3mg·l-1处理的1.75、1.37、1.37和2.33倍,且差异极显著(p<0.01)。甘草酸含量在1.81和18.1mg·l-1浓度处理时达到最大值,且两者之间差异不显著(p>0.05),与其他3个处理间存在极显著差异(p<0.01)。本研究说明适量浓度的锰处理对甘草sqs1基因的表达和甘草酸的积累具有一定的促进作用。
