采用uplc/q-tofms与nmr代谢组学技术研究生长年限对远志药材质量的影响

Abstract:

生长年限是影响远志药材质量的重要因素之一。本文对陕西合阳1~3年生栽培远志中的初级和次级代谢物进行了比较。首先对上述样品分别进行超高效液相色谱-四极杆-飞行时间质谱和核磁共振分析,采用主成分分析等多种统计分析方法,找出差异代谢物并对其含量及相关性进行分析。结果显示,1年与2、3年生远志可明显区分,而2年与3年生远志则区分不明显。随着远志生长年限的延长,初级代谢物中蔗糖、果糖、胆碱的含量减少,而甘氨酸、棉籽糖的含量则增加;次级代谢物中皂苷类成分(onjisaponinfg、polygalasaponinxxviii、polygalasaponinxxxii)的含量增加,而口山酮(polygalaxanthoneiii)及部分低聚糖酯(tenuifoliosea、tenuifoliosec、tenuifoliosec2、tenuifolioseh)的含量则减少。生长年限对远志药材质量会产生较大影响,而商品远志药材的现有生长年限有一定的科学依据。本研究为现有中药材质量评价方法提供了一种新的思路。
