萱草花总黄酮改善ccl4致大鼠肝纤维化的作用

Abstract:

本文探讨了萱草花总黄酮(hcbf)对大鼠肝纤维化的改善作用。采用ccl4构建大鼠肝纤维化模型,hcbf采用两种剂量(25、50mg·kg-1)灌胃给药,每天1次,连续50天。给药结束后,检测血清谷丙转氨酶(alt)、天冬氨酸氨基转移酶(ast)、r-谷氨酰转肽酶(ggt)、碱性磷酸酶(alp)及超氧化物歧化酶(sod)、丙二醛(mda)、转化生长因子(tgf-β1)含量;检测肝组织中i型胶原(pinp)含量;观察肝组织he染色的变化;应用免疫组化和westernblot方法检测tgf-β1蛋白表达。结果表明,hcbf组(25、50mg·kg-1)可以使alt、ast、ggt和alp水平降低(p<0.05或p<0.01),使血清中sod含量增加(p<0.01)、mda含量降低(p<0.05或p<0.01),降低tgf-β1的含量及表达(p<0.05),减少肝组织中pinp含量(p<0.05),改善肝组织的病理学改变。本研究说明,hcbf(25、50mg·kg-1)可通过减少氧化应激降低tgf-β1表达,改善ccl4引起的肝损伤。
