基于代谢组学的桑叶多组分治疗糖尿病的作用机制研究

Abstract:

采用超高效液相色谱串联静电场轨道阱高分辨质谱(uplc-orbitrapms)技术结合多变量统计学分析,利用代谢组学的研究方法和技术探讨糖尿病模型小鼠血浆整体代谢物谱特征、代谢轮廓变化和生物标志物群,并对桑叶生物碱类、黄酮类、多糖类提取物干预后的生物效应转归与分子机制进行研究。通过内源性代谢物的一级及二级碎片离子的精确分子量信息,鉴定了糖尿病模型小鼠血浆中8个潜在生物标志物,主要涉及鞘脂、氨基酸代谢途径。主成分分析表明,正常组与糖尿病模型组得到明显区分,提示模型小鼠体内代谢发生异常;桑叶生物碱类、黄酮类、多糖类提取物干预后对糖尿病模型小鼠均有不同程度的回调作用,其作用机制与其调节氨基酸代谢、鞘脂代谢密切相关。
