自噬影响衰老及老年病的研究进展

Abstract:

自噬是降解损伤及丧失功能的细胞器或蛋白,维持细胞更新和稳态的关键细胞机制。衰老是随时间逐渐出现的细胞修复机制障碍,导致损伤的分子、细胞大量堆积,并伴随功能障碍。衰老往往伴随自噬活性不足,而许多老年病受累组织也存在自噬功能异常。最近的研究证明,自噬缺失导致突变和错误折叠的蛋白质在细胞内聚集是神经退行性疾病产生和发展的物质基础,而自噬在肿瘤、组织纤维增生以及心血管等不同类型疾病或疾病的不同阶段发挥双重调节作用。本文综述了最近有关自噬参与调节衰老和几种老年病发病分子细胞机制方面的研究进展,并概述了通过调节自噬活性防治老年病的策略。
