黄芩苷-金属配合物的合成及其抗肿瘤活性研究

Abstract:
利用络合配位法合成黄芩苷-金属(ni2+、co2+、cu2+)配合物(baicalinmetalcomplexes,bmc),紫外光谱、红外光谱、质谱、热重分析、元素分析全表征bmc的组成和结构;mtt法检测bmc对smmc-7721细胞增殖的影响,pi单染法和annexin-v/fitc双染法检测bmc对smmc-7721细胞周期和凋亡的影响,荧光定量rt-pcr法检测bmc对smmc-7721细胞凋亡基因bcl-2、baxmrna的表达,流式细胞仪分析bmc对smmc-7721细胞凋亡bcl-2蛋白、bax蛋白的表达,分析bmc的抗肿瘤活性及其作用机制。结果表明,成功制备了3种新型bmc(摩尔比2:1),解析获得配合物的分子式为na2ni(c21h16o11)2·10h2o、na2co(c21h16o11)2·8h2o、na2cu(c21h16o11)2·8h2o;细胞周期和凋亡检测结果显示,bmc使细胞阻滞于g0/g1期,阻止进入s期和g2/m期;基因及蛋白检测结果显示,在设定浓度及时间条件下,bmc可使smmc-7721细胞中bcl-2基因表达下调,bcl-2表达蛋白明显降低,同时bax基因表达上调,bax表达蛋白明显升高;说明bmc通过阻滞细胞周期、下调bcl-2和上调bax的表达来抑制细胞增殖,促进细胞凋亡,3种配合物抗肿瘤活性:黄芩苷-铜(baicalin-copper,bc-cu)>黄芩苷-钴(baicalin-cobalt,bc-co)>黄芩苷-镍(baicalin-nickel,bc-ni)>黄芩苷(baicalin,bc),并呈现量-效关系。
