高硫代度红芪多糖硫酸酯的制备及体外抗凝血活性研究

Abstract:

通过氯磺酸-吡啶法对低硫代度的红芪多糖硫酸酯(shg)硫酸化修饰,控制硫酸化过程中反应温度、时间及酯化剂比例成功制备出不同硫代度的shg;紫外光谱260nm附近、红外光谱1240和810cm-1附近出现特征吸收峰,结合硫元素分析以及硫酸钡明胶比浊法共同计算出硫代度;质量浓度相同条件下随硫代度增大光谱中特征吸收峰强度也相应增强;通过体外抗凝血实验以凝血酶原时间(pt)、活化部分凝血活酶时间(aptt)、血浆凝血酶时间(tt)值来评价抗凝血活性,结果表明高硫代度的shg具有较好的体外抗凝血活性,并且在一定范围内随硫代度的提高抗凝血活性呈上升趋势。
