MT-III基因在不同海拔地区藏羊及其脑组织不同部位的差异表达研究

Abstract:

为了了解金属硫蛋白-3(Metallothionein-3,MT-III)基因在不同海拔地区饲养的藏羊脑组织及脑组织不同部位间的表达差异,随机选取高海拔地区(青海河南县)饲养的成年雄性藏羊6只和低海拔地区(甘肃临洮县)饲养的成年雄性藏羊3只,屠宰后分别在脑组织的颞叶、顶叶、延髓、小脑及松果体各部位取样,利用实时荧光定量PCR法研究MT-III基因在藏羊脑组织及其不同部位的相对表达量。结果显示,MT-III基因在藏羊脑组织各部均有表达,但不同部位相对表达量有差异;藏羊松果体中MT-III基因的相对表达量极显著高于其它部位(P<0.01);饲养在高海拔地区藏羊的脑组织的颞叶、顶叶和延髓中,MT-III基因的相对表达量均极显著高于低海拔地区饲养的藏羊(P<0.01)。MT-III是在藏羊脑组织不同部位普遍表达的一种蛋白,其在颞叶、顶叶和延髓较高的表达量可能与该区域脑细胞对低氧及高辐射的耐受性有关。
