新疆一支蒿提取物诱导棉铃虫解毒酶P450CYP6B6的表达规律

Abstract:

旨在利用实时荧光定量PCR和Westernblot分别研究新疆一枝蒿提取物黄酮及萜类对棉铃虫P450CYP6B6在mRNA水平和蛋白水平上表达的影响。实时荧光定量PCR结果表明,P450CYP6B6在mRNA水平的表达量受到诱导。时间效应为P450CYP6B6在处理的12h时能够被黄酮和萜类提取物显著诱导表达,并且在24h时表达受到抑制;浓度效应为处理24h时,黄酮和萜类提取物浓度分别为20mg/100g和1mg/100g时显著诱导P450CYP6B6的表达,并且在40mg/100g和2mg/100g时抑制P450CYP6B6的表达;Westernblot检测结果表明,萜类和黄酮在时间和浓度效应上都能显著诱导P450CYP6B6蛋白表达,并且蛋白水平在时间与浓度效应中的表达趋势与mRNA水平相似。
