LOH12CR1互作蛋白GPS2的筛选及鉴定

Abstract:

12号染色体短臂杂合子缺失的现象存在于多种血液科肿瘤和实体瘤患者中,提示此区域中可能存在抑瘤基因,LOH12CR1是此区域中已鉴定的基因之一。为探讨LOH12CR1的生物学功能,首先采取酵母双杂交系统筛查其互作蛋白,鉴定到转录调控因子GPS2是其互作蛋白。进一步在HEK293细胞中运用免疫共沉淀方法证实GPS2与LOH12CR1在体内存在相互作用;免疫荧光共定位结果显示GPS2与LOH12CR1在细胞核内存在共定位信号。
