动力学模型在荧光定量PCR数据处理中的优势

Abstract:

近年来荧光定量PCR技术凭借其基因定量的优势而得到了广泛应用,上游技术的发展保障了原始数据的精确性,但定量结果的可靠性仍取决于PCR扩增效率和所采用的数据处理模型,基于不同数学模型对原始数据处理后的结果会有较大差异。试验通过对样品进行5倍梯度稀释后获得相应梯度的荧光定量PCR原始数据,并应用2-△△Ct法、相对标准曲线法和动力学模型处理原始数据,所得定量结果(x-±s)依次为1.76±0.70、0.89±0.14和0.99975±0.06,与理论值1比较表明,动力学模型在定量结果的可靠性、方便性及经费节约方面都具有良好应用前景。
