热应激对奶山羊瘤胃黏膜紧密连接蛋白表达的影响

Abstract:

本试验旨在研究热应激对奶山羊瘤胃黏膜紧密连接蛋白表达的影响。将8只体况良好,体重、年龄、胎次和产奶量相近,处于泌乳中后期的奶山羊随机分为对照组和热应激组,每组4只羊。饲养环境采取人工控温控湿。试验期45d,在试验30和45d时,采集瘤胃背囊和腹囊组织,分别采用免疫组化法、实时定量PCR法和蛋白质免疫印迹法检测其闭合蛋白(claudin)、闭锁小带蛋白1(ZO-1)蛋白的结构分布、基因mRNA表达量和蛋白表达量。结果表明:与对照组相比,热应激组(30d)背囊和腹囊中claudin、ZO-1蛋白的信号强度和表达量均有所下降,而热应激组(45d)下降更为严重。与对照组相比,热应激组(30d)背囊和腹囊中claudin、ZO-1基因mRNA表达量均显著下降(P<0.05),而热应激组(45d)则显著低于热应激组(30d)(P<0.05)。结果提示,热应激对瘤胃黏膜的结构和完整性的破坏与瘤胃黏膜紧密连接蛋白表达量下降有关。
