果糖和胰岛素对鹅胚原代肝细胞硬脂酰辅酶A去饱和酶1基因表达量的影响

Abstract:

本试验旨在研究果糖和胰岛素对鹅胚原代肝细胞硬脂酰辅酶A去饱和酶1(SCD1)基因表达量的影响。以鹅胚原代肝细胞为试验对象,研究不同浓度果糖(0、10、30、50mmol/L)、胰岛素(0、50、100、200nmol/L)以及两者协同作用(10mmol/L果糖+50nmol/L胰岛素、10mmol/L果糖+200nmol/L胰岛素、50mmol/L果糖+50nmol/L胰岛素、50mmol/L果糖+200nmol/L胰岛素)对鹅胚原代肝细胞SCD1基因表达量的影响。结果表明:10mmol/L果糖能够极显著上调鹅胚原代肝细胞SCD1基因表达量(P<0.01),30、50、100mmol/L果糖对SCD1基因表达量均无显著影响(P>0.05);200nmol/L胰岛素能够显著上调SCD1基因表达量(P<0.05);10mmol/L果糖和200nmol/L胰岛素协同作用能极显著上调SCD1基因表达量(P<0.01)。结果提示,适宜的果糖、胰岛素单独添加以及协同作用均能上调鹅胚原代肝细胞SCD1基因表达量。
