β-防御素在金华猪和长白猪肠道中表达规律的研究

Abstract:

本试验旨在研究金华猪和长白猪从初生到60日龄不同肠段中3种β-防御素[β-防御素-1(pBD-1)、β-防御素-2(pBD-2)和β-防御素-3(pBD-3)]基因表达规律,并比较β-防御素基因在2个品种之间的表达差异。选取初生、20、40和60日龄的金华猪和长白猪各3头,屠宰后取十二指肠、空肠、回肠和结肠组织,实时定量PCR法检测各肠段pBD-1、pBD-2和pBD-3基因的表达量。结果表明:1)从初生到60日龄,随日龄的增加pBD-1和pBD-3基因在猪肠道中的表达量升高,pBD-2基因在十二指肠和空肠中的表达量升高,而在猪结肠和回肠中的表达量先下降后上升。2)在20、40和60日龄时,金华猪各肠段中3种β-防御素基因的表达量均高于长白猪。3)初生和20日龄,金华猪十二指肠pBD-1基因表达量极显著高于长白猪(P<0.01);60日龄pBD-1基因在空肠、回肠和结肠中的表达量,金华猪显著高于长白猪(P<0.05),pBD-2基因在十二指肠、回肠和结肠的表达量,金华猪显著高于长白猪(P<0.05),金华猪pBD-3基因在十二指肠、空肠和回肠的表达量显著高于长白猪(P<0.05),结肠中的表达量极显著高于长白猪(P<0.01)。结果显示,初生到60日龄,pBD-1和pBD-3基因在猪肠道中的表达呈上升趋势,金华猪肠道中3种β-防御素的表达水平在20、40和60日龄均高于长白猪。
