产油微生物的筛选

Abstract:

采用苏丹黑B染色法、甲醇-氯仿法,从30份土壤样品中筛选到9株产油霉菌和4株产油酵母.经5.8SrDNA序列分析,9株产油霉菌分别为蓝状菌Talaromycestrachyspermus、嗜松青霉Penicilliumpinophillum、青霉Pe.sp.、微紫青霉Pe.janthinellum、淡紫拟青霉Paecilomyceslilacinus、简青霉Pe.simplicissimum.、棘孢曲霉Aspergillusaculeatus、木霉Trichodermasp.、拟康宁木霉Tr.koningiopsis,4株产油酵母分别是毕赤酵母Pichiacaribbica、假丝孢酵母Candidasp.、季也蒙酵母Meyerozymaguilliermondii和季也蒙毕赤酵母Pi.guilliermondii.通过摇瓶培养,测定了各菌株的产油量,结果表明:霉菌产油量最高的是嗜松青霉,产油量为26.4%;酵母产油量最高的是假丝孢酵母,产油量为44.3%.提取毕赤酵母的油脂,采用气相色谱-质谱联用分析,测得其油脂成分主要是14~20碳的短链脂肪酸.
