绵羊卵巢腔前卵泡的分离与体外培养

Abstract:

分离方法和绵羊年龄对绵羊卵巢腔前卵泡的分离效果有显著影响.从每只羊获得腔前卵泡的总数看,机械分离法(89.5)和酶消化法(103.3)差异不显著,而正常卵泡数和正常卵泡率机械分离法(27.9,31.2%)明显高于酶消化法(19.1,18.5%);采用机械分离法,每只初情期前绵羊获得的腔前卵泡总数和正常卵泡数(125.6,35.1)均高于成年绵羊(60.,16.1),而正常卵泡率(28.1%比26.8%)差异不显著.腔前卵泡在体外培养6d,培养液中添加20ng/mL的IGF-1,培养结束时卵母细胞直径增幅为3.8μm,与对照组的29.9μm差异不显著.
