小麦黄色花叶病毒RNA2自然缺失突变体的筛选和定位分析

Abstract:

许多真菌介体传播的病毒在机械接种过程中易于发生自然缺失。利用在小麦上连续机械接种WYMV的方法,自第27代起,利用RT-PCR和Northernblot方法在感病小麦中检测到一个WYMVRNA2的缺失突变株,序列分析发现缺失区域位于RNA2的214~2808nt,共计2595nt,并在缺失区域的两端存在7个碱基的反向互补序列。缺失区域上游紧邻这7个碱基双链区存在一富含AU的短序列,并据此对此D-RNA2的缺失机制进行了讨论。
