小麦淀粉GBSSII基因的克隆、反义和RNAi载体的构建

Abstract:

采用RT-PCR方法从小麦品种豫教2号籽粒中克隆出颗粒型淀粉合成酶II基因(Granuleboundstarchsyn-thase,GBSSII)部分cDNA片段(1069bp)(GenBank登录号:EF221764),同源性比较结果显示,它与GenBank上已报道的GBSSII基因有高度的同源性。以pWM101质粒为基础,构建了由35S启动子调控的GBSSII基因的反义表达载体pWM101GBSSIIA;另外,还以pFGC5941质粒为基础,构建了GBSSII基因的RNAi载体pFGC5941GBSSIIsa,这些载体的构建为研究GBSSII基因的功能打下了很好的基础。
