基于大豆花叶病毒衣壳蛋白基因的RNA干扰植物表达载体的构建

Abstract:

大豆花叶病毒(Soybeanmosaicvirus,SMV)是马铃薯Y病毒组(Potyvirus)成员之一,大豆花叶病毒病可造成大豆10%~30%的产量损失,并严重影响大豆的品质。不同株系间致病力差异源于它们在碱基序列上的差异。采用传统的抗病育种技术育成的抗病品种抗谱窄,品种抗性可因病毒株系的变异而丢失。依靠RNA引发的基因沉默改善植物的抗病毒能力是基于RNAi(RNAinference)原理建立的植物抗病毒新策略,本研究对来源于武汉SMV分离物的CP基因和Nib基因进行了克隆,通过对保守区域的扩增,采用Gateway技术构建了大豆抗花叶病毒的RNA干扰载体。为防治大豆花叶病毒新技术的探索奠定基础。
