金华猪DGAT1、DGAT2基因发育性表达特点及其与肌内脂肪的关联分析

Abstract:

二脂酰甘油酰基转移酶是催化甘油三酯合成最后一步反应的关键酶,包括DGAT1和DGAT2两种;阐明其在发育过程中的表达规律,对于找到控制猪脂肪沉积能力的基因十分必要。运用实时荧光定量RT-PCR的方法,对60,90,120日龄金华猪肝脏、皮脂和眼肉中DGAT的mRNA表达量进行了分析,同时分析了DGAT基因mRNA表达量与肌内脂肪的相关性。结果表明,金华猪DGAT1、DGAT2基因在不同日龄的表达差异均不显著(P>0.05);在肝脏、皮脂和眼肉中,DGAT1基因表达差异均极显著(P<0.01);皮脂与肝脏、眼肉的DGAT2基因表达差异均极显著(P<0.01),而肝脏与眼肉的DGAT2基因表达差异不显著(P>0.05);而且各日龄均是皮脂中DGAT1、DGAT2基因表达量最高,肝脏次之,眼肉中最低。提示DGAT2基因在猪脂肪沉积中发挥着比DGAT1更重要的作用。研究还发现,DGAT1和DGAT2基因表达量与肌内脂肪含量无显著意义的相关(P>0.05)。
