小麦逆境胁迫相关基因TaCBL7的克隆、生物信息学及表达特性分析

Abstract:

从小麦抗性相关EST数据库中筛选并获得一条753bp的核苷酸序列,该序列包含了一个642bp的开放阅读框,编码一个由213个氨基酸残基组成的蛋白,氨基酸同源性分析发现,该蛋白含有保守的钙结合结构域EF-hand,在其N端含有豆蔻酰化位点MGXXXS/T,与OsCBL7高度同源,属于小麦CBL家族,命名为TaCBL7。实时荧光定量PCR分析表明,TaCBL7基因在苗期的根、茎及叶子,成熟期的根、茎、叶子及种子中均表达,但表达存在差异;该基因表达受盐、干旱、低温、ABA及条锈病诱导调控,表达分析结果表明,TaCBL7基因在小麦发育时间和组织空间上存在表达特异性(即时空表达特异性),同时参与了小麦生物与非生物胁迫的响应。推测TaCBL7基因编码的蛋白在小麦生长发育及胁迫过程中具有重要功能,为进一步研究小麦TaCBL7基因在生长发育和逆境响应过程中的功能,以及利用TaCBL7基因进行小麦分子育种奠定基础。
