山东地区新城疫病毒分离株的分离鉴定及其F基因的分子特性分析

Abstract:

2011年从山东不同地区的发病鸡体内分离到4株新城疫病毒,分别命名为SD1~SD4。4株毒株经病毒蚀斑克隆纯化后动物回归试验可产生典型的NDV病变|为了解其遗传起源和分子特性对其融合蛋白(Fusion,F)进行序列和分子特性的分析。研究结果发现,生物学毒力测定显示4株毒株均为强毒。F蛋白多肽裂解位点的序列为112RRQKRF117,符合NDV强毒氨基酸基序。F基因分型显示,SD01、SD02、SD03和SD04与另外一株鸡源SG01以及广东鹅分离株等属于基因Ⅶ型。同源性比较显示:SD1-SD4与同期分离株的同源性明显高于其他的传统毒株,且与国内分离株的同源性高于国外的分离株。
