AY9944-A-7对于IBMX抑制的绵羊卵母细胞体外成熟的诱导作用

Abstract:

为了探究AY9944-A-7在3-异丁基-1-甲基黄嘌呤(3-isobutyl-1-methylxanthine,IBMX)阻滞的绵羊卵母细胞减数分裂恢复过程中的作用,将绵羊卵丘-卵母细胞复合体(COCs)、分离的卵丘-卵母细胞(splitCOCs)和脱卵丘后制成的裸卵(DOs)培养于含250μmol/LIBMX的体外成熟液中,分别添加10,20,30,40μmol/L的AY9944-A-7以诱导卵母细胞恢复减数分裂。结果表明,10~40μmol/L的AY9944-A-7极显著地提高了绵羊COCs的生发泡破裂(GVBD)率和第一极体(PBI)排出率(P<0.01);添加AY9944-A-7对IBMX阻滞的绵羊DOs生发泡破裂和PBI排出没有显著的刺激作用(P>0.05);40μmol/LAY9944-A-7使绵羊COCs、splitCOCs、DOs的退化率极显著提高(P<0.01)。说明AY9944-A-7能够诱导IBMX抑制的绵羊COCs恢复减数分裂成熟,但是过量的AY9944-A-7对绵羊卵母细胞有毒性,AY9944-A-7诱导绵羊COCs克服IBMX抑制恢复减数分裂成熟的适宜剂量为20μmol/L。
