甘薯抗逆相关基因IbERF3的克隆与表达分析

Abstract:

为了提高甘薯的抗逆能力,通过RACE的方法克隆到一个含有AP2结构域的ERF家族基因,该家族基因在植物逆境调控中起重要作用。该基因cDNA全长1025bp,编码区为669bp,编码223个氨基酸,该基因在甘薯中未曾报道,将其命名为IbERF3。通过进化树分析发现,IbERF3在茄目类作物中具有一定的保守性。通过定量PCR研究发现,IbERF3在甘薯根、叶中都有表达,且在叶片中的表达量最高,同时研究发现,在干旱、盐胁迫处理后IbERF3在根和叶片中表达量都显著上升。在用植物激素ABA处理后,IbERF3的表达量逐渐上升并在24h时达到最大。因此,推测IbERF3在甘薯抗逆途径中起重要作用。
