马铃薯中VIGS沉默体系的研究

Abstract:

以马铃薯X病毒载体PVX为工具,探究不同马铃薯品种和不同侵染方式对PVX介导的VIGS沉默体系的影响,结果表明:马铃薯基因型的差异是影响VIGS沉默效果的主要因素。通过对VIGS沉默pds(八氢茄红素脱氢酶)基因的植株进行半定量RT-PCR检测,发现在所有供试的马铃薯品种中,乌盟601中pds基因的沉默效果最好,侵染后pds基因表达量下降了79.8%;而虎头、夏坡蒂和大西洋中的pds基因表达量也有不同程度降低,分别为64.1%,31.0%,4.5%。不同侵染方式即叶片注射和茎秆刺伤的试验结果表明,VIGS沉默效果和侵染方式的相关性不显著,采用叶片注射和茎秆刺伤接种马铃薯品种乌盟601,pds基因表达量分别下降了59.0%和56.5%;而虎头叶片注射和茎秆刺伤接种pds基因表达量分别下降了34.7%和40.3%。方差分析结果表明,2种不同的接种方法对pds基因沉默比例没有显著差异。
