肌动蛋白基因在棉花纤维中的作用研究

Abstract:

运用农杆菌介导法将肌动蛋白RNAi表达载体PBI121-GhACT1转化棉花,经胚胎发生获得再生植株。非转化组培再生苗作为对照。以NPTII制备探针,Southern杂交结果表明,获得8个独立遗传转化系,其中个系有2个拷贝插入;其余6个系为多拷贝。显微镜下观察开花后第5天的幼胚珠孔端,对照纤维长度约是RNAi载体转基因的~5倍。成熟后绒长F测验结果表明,RNAi载体转化株与对照差异显著,RNAi转基因植株的绒长显著短于对照。说明GhACT1基因对纤维有延缓和抑制作用,并且主要作用于纤维发育早期。
