辽宁绒山羊SRY基因编码区的分子特征研究

Abstract:

根据GenBank中山羊SRY基因序列设计一对引物,采用PCR在雄性辽宁绒山羊个体中扩增出了包含SRY基因整个编码区的特异片段,将特异PCR产物克隆到pMD18-T载体中,转化DHα菌,筛选出SRY基因的阳性克隆,进行了测序,将其序列与GenBank中部分偶蹄目动物的SRY基因序列进行同源性比较,用NJ法构建了系统进化树。结果表明,辽宁绒山羊SRY基因编码区长度为723bp,共编码240个氨基酸,其中包含长度为231bp的HMG-box,位于编码区的第190至420位核苷酸之间,编码77个氨基酸(H64-A140)。辽宁绒山羊SRY基因编码区序列与GenBank中山羊、绵羊、牛、牦牛、水牛、梅花鹿、麝香鹿及猪等偶蹄目动物的同源性分别为100%,97.09%,89.21%,89.21%,88.38%,87.%,86.16%和68.04%;基于SRY基因编码区的核苷酸序列,构建的物种间系统树结果基本上与这些偶蹄目动物的传统系统分类相一致。
