片球菌素基因的克隆及表达

Abstract:

从乳酸片球菌中提取质粒DNA作为模板,根据GeneBank公布的细菌素结构基因,设计一对特异性引物,进行PCR扩增片段,将该片段定向克隆到pTA2载体,测序,与发表的基因序列比较,同源性为100%,然后克隆到表达载体Pet-28a,转化到EscherichiacoliDH5α,经过卡那霉素平皿筛选,质粒酶切,PCR两步验证,获得阳性重组质粒,并转化表达宿主菌E.coliBL21(DE3)感受态细胞,以异丙基硫代-βD-半乳糖苷(IPTG)诱导表达,Tricine-SDS-PAGE电泳,显示在4600Da处出现条带,与已报道的细菌素分子量大小符合.表达产物对单核细胞增多症李氏杆菌有抗菌作用.
