我国长江流域PRRSVORF3基因的遗传变异分析

Abstract:

为了确定猪繁殖与呼吸综合征病毒(PRRSV)的流行特点及变异规律,本研究首次对从200年以来我国长江流域分离到的29株PRRSV的ORF3基因进行了克隆和序列测定,并与国外代表毒株和国内的高致病性PRRSV进行了比较分析.结果表明:所有分离毒株的ORF3基因均编码254个氨基酸,都属于美洲型PRRSV,推导的氨基酸同源性为84.%~100%,与美洲型PRRSVVR-2332的同源性为85.0%~97.2%;与国内分离的高致病性PRRSV的同源性为8.%~100%.根据ORF3基因的遗传进化分析,这些毒株可分为两个亚群,毒株HZ24与VR-2332及疫苗株亲缘关系较近,其他分离株与高致病性PRRSV处在同一进化分支.同时,该基因编码蛋白的抗原表位和抗原性也发生了变化.
