强雌性系西瓜ACC合成酶基因的克隆及其序列分析

Abstract:

为了解ACC合成酶基因对西瓜花性型分化的作用机理,根据已报道的西瓜ACC合成酶(ACS)基因序列,设计特异性引物,以强雌性西瓜总RNA为模板,通过RT-PCR扩增得到4条特异性片段:Cit-ACS1为1690bp、Cit-ACS2为402bp、Cit-ACS3为598bp和Cit-ACS4为499bp。序列同源性比对表明,该4个片段与已报道的西瓜ACC合成酶基因序列的同源性99.8%~100%。构建了西瓜ACC合成酶基因系统进化树,显示Cit-ACS1与甜瓜CMe-ACS2和黄瓜CS-ACS2亲缘关系最近;并对Cit-ACS1基因编码的蛋白进行了二级结构和三级结构分析。
