人工异源六倍体小麦中甲基化差异的MSAP分析

Abstract:

利用MSAP方法分析了小麦从四倍体到六倍体进化过程中甲基化水平和甲基化遗传模式的变化。结果表明,人工异源六倍体小麦SCA/SQ(AABBDD)及其四倍体母本SCAUP(AABB)、二倍体父本粗山羊草SQ523(DD)的总甲基化水平分别为28.21%,27.53%,24.11%。37对引物检测到1087条差异的扩增片段,对这些位点的甲基化遗传模式进行分析,结果表明,在人工异源六倍体中发生过甲基化的比例为18.95%,高于去甲基化的比例(2.58%)。这些结果揭示了小麦从四倍体到六倍体的进化过程中,通过对一些功能基因的甲基化来减轻异源多倍化造成的影响。
