短乳杆菌产NADH氧化酶的发酵及纯化

Abstract:

有氧环境下,利用优化后的发酵培养基,研究短乳杆菌(Lactobacillusbrevis)产NADH氧化酶(NOX)的发酵条件.采用8层纱布,在温度为30℃,pH=7.0,转速为150r.min-1的条件下发酵培养23h,用pH值为5.8的0.1mol.L-1磷酸钾缓冲液对细胞进行洗涤悬浮,超声波破碎(功率为400W,超声2s,间歇2s,破碎12min)后,测得其比活力可达2.317mkat.g-1.通过DEAESepharoseFastFlow,Macro-PrepHighQ和Hydroxyapatite-Ⅰ三步纯化后,其纯化倍数为3.11,回收率为8.50%.
