生物酶降解米糠释放阿魏酸工艺条件优化

Abstract:

采用均匀设计法设计和二次多项式逐步回归分析,优化一株放线菌降解米糠释放反式阿魏酸的培养基配方,并采用单因素法优化发酵条件.优化后培养基配方:米糠质量分数为6%,米糠粒径0.50mm,酵母粉质量分数为1.0%; 优化后的发酵条件:发酵温度为28℃,摇床转速200r.min-1,装液系数为12%,接种量的体积分数为3.3%,pH值8.0.实验结果表明,反式阿魏酸的释放率最高为14.55%,与优化前相比,释放率提高了106.7%.研究阿魏酸酯酶和木聚糖酶联合作用于米糠产阿魏酸和低聚糖的协同效应,阿魏酸释放率达到20.22%,与阿魏酸酯酶单独降解米糠相比,其释放率提高了119.1%; 低聚木糖的释放量2.04mg.mL-1,与木聚糖酶单独降解米糠相比,其释放量提高了87.2%.
