濒危植物延龄草初代培养中无菌培养物的建立

Abstract:

延龄草为国家三级保护植物,近几年,其资源数量急剧减少,急需寻找增加资源的途径.为了获得延龄草初代培养的无菌培养物,对延龄草初代培养中外植体所感染的微生物进行分离纯化、形态学及生理生化鉴定,进一步对污染外植体进行抑菌处理.结果表明,外植体所感染菌株均为埃希氏菌属(Escherichia);在培养基中加200mg/L链霉素或300mg/Lα-氨苄青霉素能获得较好的抑菌效果;100mg/L链霉素或150mg/Lα-氨苄青霉素浸泡污染外植体6h也能获得较好的抑菌效果.该研究为延龄草离体繁殖体系的建立奠定了基础,从而为进一步从离体繁殖的角度扩大资源提供了可能.
