合成的西红花糖苷体外抗肿瘤活性研究

Abstract:

西红花提取物(saffronextract,SE)具有潜在的抗肿瘤活性.通过以下方法比对SE及合成的西红花糖苷体外抗肿瘤活性的差异并由此分析后者抗肿瘤机制MTT法比较肿瘤细胞增殖活性,流式细胞术检测合成的西红花糖苷处理后肿瘤细胞的凋亡及其细胞周期的变化,Westernblot法探究合成的西红花糖苷对p53基因表达的影响.实验发现,与天然SE比较,合成crocetin、crocinI~crocinIII体外抑制肿瘤细胞增殖的能力增强,而HCT116、A549两种细胞对其较为敏感;另外,与空白对照比较,合成的crocetin、crocinI~crocinIV可诱导肿瘤细胞凋亡、阻止细胞周期进程,其机制可能与促进p53基因表达相关.本研究部分揭示了合成的西红花糖苷的体外抗肿瘤活性,并对相关机制进行了一定的探索,为探寻更理想的西红花类抗肿瘤药物提供了体外研究的依据.
