内皮素-1反义脱氧寡核苷酸对糖尿病大鼠心肌超微结构的影响

Abstract:

目的研究内皮素-1反义脱氧寡核苷酸(ET-1AS-ODN)对链脲佐菌素(streptozotocin,STZ)诱导的糖尿病大鼠心肌超微结构的影响。方法一次性腹腔注射STZ(60mg?kg-1),建立大鼠糖尿病模型,并饲养5周后,造成糖尿病心肌损伤大鼠模型,ET-1AS-ODN干预,采用BL-420生物信号采集系统检测大鼠心功能,透射电镜观察大鼠心肌超微结构。结果ET-1AS-ODN各剂量组均可使LVEDP、+dp/dtmax、-dp/dtmax表现出一定的改善(P<0.05,P<0.01);电镜观察可见ET-1AS-ODN各剂量组心肌细胞超微结构均明显改善,表现心肌细胞内肌节结构清晰、排列基本正常,线粒体增多、肿胀减轻、内嵴结构趋于清晰,排列整齐。结论内皮素-1反义寡核苷酸可减轻STZ诱导的糖尿病大鼠的心肌损伤,对心肌组织具有保护作用。
