缺血预处理大鼠心肌组织BDNF表达及其意义

Abstract:

目的观察缺血预处理大鼠心肌中脑源性神经生长因子(BDNF)的mRNA及蛋白表达情况,并探讨其意义。方法54只雄性SD大鼠随机分为空白对照组(Con组)、缺血再灌注组(I/R组)和缺血预处理组(IPC组),I/R组及IPC组模型建立后,再分别分为再灌注后1、3、6h及12h各时间点亚组。RT-PCR检测心肌组织BDNFmRNA表达,Western-blot法检测心肌组织BDNF蛋白表达。结果I/R组及IPC组各时间点大鼠心肌组织中BDNFmRNA及蛋白的表达与Con组相比,差异均有统计学意义(P<0.05);而I/R组与IPC组再灌注后同时间点比较大鼠心肌组织中BDNFmRNA及蛋白表达的差异也均有统计学意义(P<0.05)。结论缺血预处理可以上调BDNF的mRNA及蛋白表达,推测BDNF在缺血预处理后对缺血再灌注大鼠心肌具有保护作用。
