不同病程糖尿病大鼠视网膜病变的研究

Abstract:

目的观察不同时间点糖尿病大鼠视网膜病理结构及功能的改变,探讨糖尿病大鼠视网膜病理变化与病程的关系。方法取健康雄性SD大鼠64只,随机分为正常对照组(CON组),糖尿病1个月组(DM1组)、3个月组(DM2组)、5个月组(DM3组),糖尿病组实验前用链脲佐菌素(STZ)制造糖尿病大鼠模型。随机抽取相应DM1、DM2、DM3组及正常对照组大鼠,用透射电镜观察不同时间点的糖尿病大鼠和正常大鼠视网膜的超微结构,视网膜HE染色切片、伊文思蓝(EB)视网膜铺片及视网膜EB含量测定观察视网膜结构及功能的变化。结果病程初期糖尿病大鼠视网膜色素上皮细胞、膜盘、外核层、内核层及神经节细胞等超微结构已出现改变,且病变随病程进展而加重。视网膜HE染色示CON组视网膜未见明显异常,DM1、DM2、DM3组视网膜各层组织病理改变随病程延长而加重。EB染色视网膜铺片显示于糖尿病3个月时已出现血管荧光渗漏,5个月时视网膜血管出现结构明显不规则和大量荧光渗漏。视网膜EB含量随糖尿病病变加重而不断增多,CON组、DM1组、DM2组、DM3组视网膜EB含量分别为(0.227±0.051)、(0.325±0.054)、(0.510±0.027)、(0.721±0.035)ng?mg-1,差异有统计学意义(P<0.05)。结论糖尿病大鼠视网膜结构在1个月已出现病变,且随病程延长而加重。
