重组干扰素-α增强口蹄疫重组疫苗免疫效果的研究

Abstract:

目的观察重组干扰素-a(IFN-α)佐剂对口蹄疫(FMD)重组疫苗免疫效果的增强作用,为IFN-α作为疫苗佐剂应用提供实验依据。方法6周龄雌性BALB/c小鼠,分为3组,实验组联合免疫表达猪IFN-α的重组腺病毒(rAd5poIFN-α)和串联表达FMDV多表位的重组腺病毒(rAd5EGS),同时设两组对照组,分别注射rAd5EGS和PBS,间隔两周免疫1次,共免疫3次。首免后每周分离血清,并通过ELISA检测血清中抗FMDVVP1特异性IgG;首免后6周检测血清中IgG1、IgG2a抗体亚型及细胞因子IL-4、IFN-γ的表达水平;同时分离脾细胞,MTT法检测淋巴细胞增殖指数。结果与单独免疫rAd5EGS和注射PBS的对照组相比,联合免疫rAd5poIFN-α和rAd5EGS能明显增强特异性IgG的分泌和小鼠淋巴细胞增殖功能,并能增强抗体亚型IgG2a和细胞因子IFN-γ的分泌,但对抗体亚型IgG1和IL-4的分泌没有明显的增强作用。结论重组腺病毒rAd5poIFN-α能有效增强FMD重组疫苗诱导的Th1型细胞免疫,有望作为FMD重组疫苗的候选佐剂。
