枸杞多糖对大剂量化疗后小鼠睾丸c-kit基因表达的影响

Abstract:

目的探讨枸杞多糖(Lyciumbarbarumpolysaccharides,LBP)对大剂量化疗后小鼠睾丸c-kit基因表达的影响。方法4-6周龄雄性昆明白(KM)小鼠150只,随机取其中10只作为正常对照组,其余140只腹腔内单次注射40mg?kg-1的白消安建立大剂量化疗致雄性小鼠不育模型。将40只造模成功的小鼠随机分为化疗+生理盐水组、化疗+50mg?(kg?d)-1LBP组、化疗+100mg?(kg?d)-1LBP组、化疗+200mg?(kg?d)-1LBP组,每组10只,LBP灌胃。观察动物的一般状况,HE染色观察各实验组睾丸组织结构,实时荧光定量PCR检测分析LBP对各实验组小鼠睾丸c-kitmRNA表达的影响。结果HE染色观察发现模型组小鼠曲细精管内细胞层数减少,多数仅存支持细胞,管腔呈空泡样变;LBP治疗组小鼠曲细精管管腔空巢率减少,重现精子。实时荧光定量PCR检测结果正常对照组、化疗+生理盐水组、化疗+50mg?(kg?d)-1LBP组、化疗+100mg?(kg?d)-1LBP组、化疗+200mg?(kg?d)-1LBP组的c-kitmRNA相对表达量分别为(15.27±0.13)、(5.94±0.11)、(8.58±0.27)、(15.05±0.35)、(15.06±0.21)。模型组(化疗+生理盐水组)较正常对照组c-kitmRNA的表达有明显下降,差异有统计学意义(P0.05)。结论LBP可以显著提高大剂量化疗后小鼠睾丸c-kit基因的表达,100mg?(kg?d)-1可能是LBP对大剂量化疗后小鼠睾丸生精功能恢复的最佳起始剂量。
