IKKα-siRNA可抑制SKOV3细胞的生长

Abstract:

目的研究IKKα在卵巢癌组织中的表达情况及其对卵巢癌细胞生长增殖能力的影响,探讨IKKα在卵巢癌细胞发生发展中的作用。方法采用实时定量PCR方法检测人卵巢癌组织中IKKα的表达水平,设计并合成针对IKKα的siRNA,利用脂质体转染法将其转入人卵巢癌细胞SKOV3中,通过MTT和平板克隆形成实验观察IKKα-siRNA对SKOV3细胞生长活性和增殖能力的影响。结果实时定量PCR结果显示,IKKα在卵巢癌组织中的表达水平明显高于正常卵巢组织(P<0.05)。MTT和平板克隆形成实验表明,转染IKKα-siRNA的SKOV3细胞的生长活性和集落形成率明显低于对照组(P<0.05或P<0.01)。结论IKKα在卵巢癌组织中的表达水平显著上升,IKKα-siRNA可抑制SKOV3细胞的生长活性和增殖能力,提示IKKα可能与卵巢癌的恶性发生、发展进程相关。
