大鼠异基因骨髓移植模型的实验研究

Abstract:

目的本研究拟通过全身照射,采取Y染色体标记,通过全骨髓细胞移植,建立SD大鼠性别错配异基因骨髓移植模型,并探讨建立该模型的可行性及其意义,为干细胞示踪等研究提供实验参照。方法以雄性SD大鼠为供鼠,雌性SD大鼠为受鼠,受体大鼠随机分成移植组和对照组,每组各10只。移植当天移植组和对照组大鼠均接受8.0Gy的全身照射,于照射后6h,移植组经鼠尾静脉输入2×108个/kg的雄性供鼠全骨髓细胞;对照组组注射等量的RPMI1640培养液。观察两组大鼠存活情况,检测移植组大鼠生存期外周白细胞计数,采用PCR技术检测SRY基因。结果采用8.0Gy的全身照射为预处理方案,对照组大鼠2周内全部死亡,而移植组大鼠30d的存活率为70%。外周血细胞计数提示全骨髓移植后造血功能恢复,且检测有雄性供鼠Y染色体出现,提示SD大鼠性别错配异基因骨髓移植成功。结论应用8.0Gy的全身照射加供体骨髓移植可成功建立Y染色体标记的同种异体大鼠嵌合体模型。
