损伤的鼠心肌细胞cTnI和LDH释放浓度和时间关系

Abstract:

目的通过测定培养两天龄大鼠心肌细胞释放的完整肌钙蛋白I(cTnI)分子和乳酸脱氢酶(LDH)的释放,探讨二者释放浓度与时间的关系。方法分别用ELISA法和丙酮酸显色法测定体外培养的两天龄大鼠心肌细胞在不同浓度代谢抑制剂(NaCN)和Ca2+负荷及不同作用时间下cTnI和LDH的释放量。结果与LDH释放相比,相同代谢抑制剂浓度和细胞外Ca2+浓度下,cTnI的释放明显晚于LDH,LDH与cTnI的释放量有显著差异(P<0.01);cTnI的释放是缓慢的、部分的,cTnI的释放量与代谢抑制剂浓度或细胞外Ca2+呈正相关。结论在化学性缺氧和Ca2+负荷下,完整cTnI分子从受损及死亡的心肌细胞内释放;完整cTnI分子的释放量是心肌细胞重度损伤、死亡的指标,但不是早期指标。
