缝隙连接蛋白43在缺血预处理心肌保护中的作用

Abstract:

目的探讨缝隙连接蛋白43(Connexin43,Cx43)在缺血预处理心肌保护中的作用和地位。方法将48只SD大鼠分为假手术组(S组)、缺血再灌注组(IR组)、缺血预处理组(IP组)和5-羟基癸酸+缺血预处理组(5-HD+IP组),测定各组的心律失常评分和心肌梗死面积,采用逆转录-聚合酶链反应技术(RT-PCR)检测各组心肌Cx43的表达水平。结果IR组和5-HD+IP组的心律失常发生多于IP组,评分明显高于IP组(P<0.01);IR组和5-HD+IP组的IS/AAR比值明显大于IP组(P<0.01);与IR组和5-HD+IP组比较,IP组Cx43mRNA的表达明显增强(P<0.01)。IP可以减少心律失常发生,减小心肌梗死面积,增强Cx43的表达,5-羟基癸酸可以阻断IP的心肌保护作用。结论IP通过开放线粒体ATP敏感性钾通道保持Cx43磷酸化,进而实现心肌保护。
